  • C-peptide, historically considered a byproduct of insulin maturation, is now recognized for potential physiological roles and serves as a clinically important biomarker of endogenous insulin secretion.
  • The protein is expressed and secreted by pancreatic beta-cells, with tissue-specific detection at the protein level in these cells.

  • The immunogen sequence (amino acids 1-100 of human INS) encompasses the B-chain and most of C-peptide; therefore, the antibody may recognize proinsulin and partially processed intermediates, but not mature insulin (which lacks the C-peptide region). For specific C-peptide detection, validate against proinsulin cross-reactivity using appropriate controls.
  • For ELISA applications, consider using the antibody in a capture or detection format after confirming linearity and specificity with recombinant C-peptide and proinsulin standards in your sample matrix.
  • In immunohistochemistry (IHC-P), paraffin-embedded pancreatic tissue sections should show strong staining in islet beta-cells; antigen retrieval may be required for optimal signal.
  • For immunofluorescence (IF/ICC and IF-P), permeabilization is recommended to access intracellular C-peptide stores; include an isotype control to assess background.
