Biological Background: Complement C4-A Function and Localization

  • Complement C4-A is a non-enzymatic component of the classical, lectin, and GZMK complement pathways, serving as a precursor that is proteolytically activated to participate in innate immune defense.
  • Upon cleavage by complement proteases such as C1S, MASP2, or GZMK, it generates C4b, which covalently attaches to pathogen surfaces, acting as an opsonin to mark antigens for removal. Related references: PMID:8538770 PMID:27738201
  • C4b then recruits complement C2b to form the C3 and C5 convertases, which cleave and activate C3 and C5, propagating the complement cascade. Related references: PMID:12878586 PMID:18204047 PMID:2387864
  • The C4a fragment, released during activation, is structurally similar to anaphylatoxins; however, its role remains unclear and earlier reports of inflammatory mediator function may be due to contamination. Related references: PMID:6167582 PMID:25659340

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a C-terminal fragment (aa 940-1237), which may confer isoform-specificity; however, cross-reactivity with the highly homologous C4B should be experimentally verified.
  • For Western blotting, use low-percentage polyacrylamide gels and extended transfer times to efficiently resolve and transfer the high molecular weight target (~193 kDa).
  • In immunofluorescence (IF-P), since C4A is secreted and can be deposited on cell surfaces, optimize fixation and permeabilization conditions to detect both intracellular and surface-bound protein.
  • For ELISA, this antibody can be used as a capture or detection reagent; pair with another clone targeting a non-overlapping epitope for sandwich ELISA development.
  • As complement activation can consume C4, consider sample handling and storage conditions to preserve native protein levels.
