Biological Background: Complement C3 Function and Localization

• C3 is a ~187 kDa precursor protein that is post-translationally cleaved into functional fragments C3a (anaphylatoxin) and C3b (opsonin). The C3b fragment contains a reactive thioester bond enabling covalent attachment to pathogen surfaces.

• Since C3 is a secreted protein, consider fixation and permeabilization if intracellular staining of C3-producing cells (e.g., hepatocytes, adipocytes) is required. For surface detection of C3b deposits on target cells, no permeabilization is necessary.
• Validate the assay in your specific sample system, such as human whole blood, peripheral blood mononuclear cells (PBMCs), or purified cell populations, and confirm cross-reactivity is restricted to human C3 as indicated.
