Biological Background: p53 Function and Localization
- Multifunctional transcription factor that acts as a tumor suppressor by inducing cell cycle arrest or apoptosis upon DNA damage. Functions by binding to specific DNA sequences to regulate target gene expression. Related references: PMID:11025664, PMID:12524540, PMID:12810724
- Mediates apoptosis partly through transcriptional activation of pro-apoptotic genes such as BAX and FAS, and repression of anti-apoptotic Bcl-2. - For Western blotting, treatment of cells with DNA-damaging agents (e.g., doxorubicin or UV irradiation) can increase p53 phosphorylation at S376, providing a positive control. The predicted molecular weight is approximately 44 kDa, but multiple bands may appear due to splice isoforms.
- In ELISA, consider using recombinant p53 protein phosphorylated at S376 as a standard and validate assay specificity with non-phosphorylated peptide controls.
- Cross-reactivity has been confirmed with human and rat p53; for other species, sequence homology analysis around S376 is advised before use.
