Biological Background: p53 Function and Localization

  • p53 is a multifunctional transcription factor and tumor suppressor, inducing cell cycle arrest, DNA repair, or apoptosis upon binding to its target DNA sequence (UniProt P04637). Related references: PMID:11025664, PMID:12524540, PMID:12810724
  • It acts as a tumor suppressor in many tumor types, negatively regulating cell division by controlling expression of genes required for this process. Related references: PMID:11025664, PMID:12524540, PMID:12810724
  • Apoptosis induction by p53 is mediated via stimulation of BAX and FAS antigen expression, or by repression of Bcl-2 expression (PubMed:12524540, PubMed:17189187). Related references: PMID:12524540, PMID:17189187
  • p53 localizes to multiple subcellular compartments, including the nucleus, cytoplasm, nucleoplasm, PML bodies, endoplasmic reticulum, mitochondrial matrix, and centrosome.
  • Multiple isoforms of p53 are expressed in a tissue-dependent manner; some isoforms enhance or suppress the transactivation activity of isoform 1.
  • Key post-translational modifications of p53 include acetylation, phosphorylation, methylation, and ubiquitination (Ubl conjugation), as indicated by UniProt keywords.
  • p53 is also involved in regulating the circadian clock by repressing CLOCK-BMAL1-mediated transcriptional activation of PER2 (PubMed:24051492). Related references: PMID:24051492

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant human p53 protein; this may suggest the antibody can recognize both native and denatured p53, but experimental validation is recommended.
  • For Western blotting, a band at approximately 44 kDa is expected; use appropriate positive (e.g., HEK293, HeLa) and negative (p53 knockout) cell lysates to confirm specificity.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization conditions, and include isotype controls.
  • ELISA applications may require coating with recombinant p53 or cell lysates; validate the antibody in the intended assay system.
  • The antibody is KO validated; consider using KO cell lines or tissues as essential controls to ensure signal specificity.
