- The immunogen is a synthetic phosphopeptide corresponding to residues surrounding Ser33 of human p53. To validate phospho-specificity, consider using lambda phosphatase-treated samples or non-phosphopeptide competition in parallel.
- For western blot (WB), predicted band size is ~53 kDa (canonical p53) or 44 kDa (some isoforms); bands may shift slightly due to phosphorylation and other modifications.
- For immunohistochemistry (IHC-P), antigen retrieval conditions and tissue fixation can affect phospho-epitope detection; optimize using known positive controls such as irradiated or genotoxic-stress treated tissues.
- When performing immunoprecipitation (IP) or ELISA, include appropriate IgG controls and consider pairing with a total p53 antibody to assess phosphorylation stoichiometry.
