Biological Background: Histone H4 Function and Localization
Histone H4 is a core component of the nucleosome, around which DNA is wrapped to form chromatin. This fundamental structure compacts genomic DNA and regulates its accessibility to enzymes and other factors required for DNA-templated processes such as transcription, replication, and repair. Histones, including H4, are therefore central to maintaining chromosomal stability and epigenetic control.
A complex set of post-translational modifications, often referred to as the histone code, regulates chromatin dynamics. Histone H4 undergoes numerous modifications, including acetylation, methylation, phosphorylation, and citrullination, at various residues along its N-terminal tail and globular domain.
This antibody specifically targets the asymmetric dimethylation of arginine 3 (R3) in the N-terminal tail of histone H4. In the processed protein, this corresponds to residue 4 (R4). This modification is catalyzed primarily by protein arginine methyltransferase 1 (PRMT1) and plays a role in transcriptional activation and chromatin remodeling.

Histone H4 is localized to the nucleus and chromosomes. During spermatogenesis, acetylation of H4 is associated with its nuclear localization in step 11 spermatids.
