Biological Background: Histone H3 (Asymmetric Dimethyl-R8) Function and Localization
Histone H3 is a core component of the nucleosome, which wraps and compacts DNA into chromatin, limiting DNA accessibility to cellular machineries. Histones play a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is dynamically regulated through a complex set of post-translational modifications (PTMs), collectively known as the histone code, and by nucleosome remodeling. Among the numerous PTMs on histone H3, arginine methylation is catalyzed by protein arginine methyltransferases (PRMTs) and can occur in asymmetric or symmetric forms. Asymmetric dimethylation of H3R2 is mediated by PRMT6 PMID:17898714, while asymmetric dimethylation of H3R17 is catalyzed by CARM1 PMID:15345777. The H3R8 residue is known to be symmetrically dimethylated by PRMT5; however, asymmetric dimethyl-H3R8 has also been reported as a specific epigenetic mark. This antibody targets the asymmetric dimethylation state at R8, enabling discrimination of this modification.
Histone H3 is localized to the nucleus and chromatin. The canonical H3.1 isoform (P68431) is expressed ubiquitously, whereas the H3.1t variant (Q16695) is specifically expressed in testicular cells. Related references: PMID:16567635 (citrullination at R8/R2/R17)
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide bearing asymmetric dimethylation at R8; the antibody is expected to specifically recognize this modification. However, due to sequence homology, cross-reactivity with other methylated histone H3 variants or sites may occur. Validate specificity using peptide competition, knockout/knockdown, or site-directed mutagenesis.
- For Western blot, the expected molecular weight is ~15 kDa. Histones are highly basic; use acid-urea gels or include appropriate controls to resolve modified forms.
- For ChIP, consider that asymmetric dimethyl-H3R8 may mark specific chromatin states; optimize crosslinking and shearing conditions.
- Applications listed include WB, IHC-P, IF/ICC, ELISA, and ChIP. Dilutions should be empirically determined, and all assays should include appropriate negative and positive controls.
- Cross-reactivity is predicted against human, mouse, rat, and other species due to high conservation of histone H3. Verify in the relevant sample system.
