Experimental Guidance and Technical Tips

  • This antibody was raised against a synthetic peptide spanning amino acids 1-100 of human Histone H4 (NP_003529.1), with symmetric dimethylation at Arg3. The immunogen design suggests specificity for the modified form, but cross-reactivity with unmodified or other modified forms should be assessed.
  • For Western blotting (WB), the predicted molecular weight of Histone H4 is ~11 kDa. Consider using nuclear extracts or histone-enriched preparations as positive controls. Reduce and denature samples prior to loading.
  • In immunohistochemistry (IHC) and immunofluorescence (IF/ICC), antigen retrieval may be required to expose the epitope, especially in formalin-fixed, paraffin-embedded tissues. Test multiple antigen retrieval methods.
  • Validate the antibody specificity by including appropriate controls, such as peptide competition assays or comparison with cells treated with methyltransferase inhibitors.
