Biological Background: Histone H4 Function and Localization
- Histone H4 is one of the core histones that form the nucleosome, the fundamental unit of chromatin. Together with histones H2A, H2B, and H3, an octamer of histones wraps ~147 bp of DNA, compacting it and regulating access to the genetic material.
- This packaging is essential for DNA-templated processes including transcription, replication, repair, and maintaining chromosomal stability. The dynamic regulation of chromatin structure is governed by post-translational modifications (PTMs) on histones, often referred to as the “histone code”.
- Histone H4 is primarily located in the nucleus and is associated with chromosomes. It can be localized to the nucleus when specific modifications occur, e.g., acetylation during spermatogenesis.
- H4 undergoes a vast array of PTMs, including acetylation, methylation, phosphorylation, citrullination, and various acylations. These modifications occur at multiple residues and regulate distinct downstream events.
- The symmetric dimethylation of arginine 3 (H4R3me2s) is catalyzed by protein arginine methyltransferases PRMT5 and PRMT7. This modification is implicated in transcriptional regulation and is one of the many marks that comprise the histone code. Related references: UniProt P62805.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic peptide spanning amino acids 1-100 of human Histone H4 (NP_003529.1), with symmetric dimethylation at Arg3. The immunogen design suggests specificity for the modified form, but cross-reactivity with unmodified or other modified forms should be assessed.
- For Western blotting (WB), the predicted molecular weight of Histone H4 is ~11 kDa. Consider using nuclear extracts or histone-enriched preparations as positive controls. Reduce and denature samples prior to loading.
- In immunohistochemistry (IHC) and immunofluorescence (IF/ICC), antigen retrieval may be required to expose the epitope, especially in formalin-fixed, paraffin-embedded tissues. Test multiple antigen retrieval methods.
- Validate the antibody specificity by including appropriate controls, such as peptide competition assays or comparison with cells treated with methyltransferase inhibitors.
- The antibody is predicted to react with a wide range of species due to high sequence conservation of Histone H4; however, verify reactivity in your specific model system.
- Optimize dilution factors empirically for each application; typical starting dilutions may be adapted from existing protocols for polyclonal antibodies targeting histone modifications.
