Biological Background: Histone H3 Function and Localization

Histone H3 is a core component of the nucleosome, the fundamental unit of chromatin. Nucleosomes wrap and compact DNA, limiting accessibility to the cellular machinery and thereby playing a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability.

Histone H3 is subject to a vast array of post-translational modifications (PTMs), including acetylation, methylation, phosphorylation, citrullination, and many others, collectively referred to as the "histone code." These modifications regulate chromatin structure and gene expression dynamically. The N-terminal tail of histone H3 is particularly rich in modifiable residues, with lysine 27 (K27) being a critical site for methylation events. Di-methylation of H3K27 (H3K27me2) is associated with transcriptional repression and heterochromatin maintenance.

The human genome encodes several histone H3 variants. Q16695 (H3-4, also known as H3.1t) is a testis-specific histone primarily expressed in testicular cells, while P68431 (H3C1/H3C2/H3C3/H3C4/H3C6/H3C7/H3C8/H3C10/H3C11/H3C12) represents the canonical replication-dependent H3.1 variants expressed in somatic cells. Both variants are highly conserved and share identical N-terminal sequences where K27 is located. They localize exclusively to the nucleus and chromosomes, where they form an integral part of chromatin fibers.

Experimental Guidance and Technical Tips

  • For Western blot (WB), a single band around 15 kDa corresponding to histone H3 is expected. Use appropriate positive and negative controls to confirm band identity and dimethylation specificity.
  • For ChIP and ChIP-seq, optimize cross-linking (e.g., formaldehyde) and chromatin shearing conditions. Consider validating enrichment of known H3K27me2 target loci using qPCR or parallel sequencing.
  • The antibody cross-reacts with human, mouse, and rat, and is predicted to have a wide range of reactivity. However, it is advisable to validate performance in the specific species and tissue of interest before large-scale experiments.
  • For ELISA and dot blot (DB), titrate the antibody and include blocking steps to minimize non-specific binding; the peptide immunogen can serve as a positive control.
  • In immunohistochemistry (IHC) and immunofluorescence (IF/ICC), note that histone H3 is nuclear; a counterstain (e.g., DAPI) can help visualize nuclear localization. Antigen retrieval may be necessary.
