Biological Background: Histone H3 Function and Localization

Histone H3 is a core component of the nucleosome, wrapping and compacting DNA into chromatin to limit accessibility for transcription, replication, and repair. Through a complex array of post-translational modifications (PTMs), histones play a central role in the regulation of gene expression, DNA repair, DNA replication, and chromosomal stability—a phenomenon often referred to as the histone code.

Asymmetric dimethylation of arginine residues (Rme2a) on histone tails, including at R8 of Histone H3, is catalyzed by protein arginine methyltransferases (PRMTs) and contributes to transcriptional activation or silencing depending on the genomic context. This specific modification is part of a broader epigenetic landscape that governs chromatin dynamics.

Two major Histone H3 variants are targeted by this antibody: Histone H3.1t (encoded by H3-4/HIST3H3, UniProt Q16695) and the canonical Histone H3.1 (encoded by multiple H3C genes, UniProt P68431). While Histone H3.1 is widely expressed, Histone H3.1t shows testis-specific expression. Both variants localize to the nucleus and chromosome.

Experimental Guidance and Technical Tips

  • Western Blot (WB): A single band around 15 kDa is expected. Use nuclear extracts or acid-extracted histones as positive controls. Consider including unmodified peptide competition to confirm specificity for the asymmetric dimethyl-R8 modification.
  • Dot Blot (DB): Spot peptide antigens (asymmetric dimethyl-R8 vs. unmodified or symmetric dimethyl-R8) to assess antibody specificity.
  • Immunohistochemistry (IHC-P): Antigen retrieval with citrate buffer (pH 6.0) is recommended. Validate signal in tissues known to express H3R8me2a, and use isotype controls to confirm staining specificity.
  • ELISA: Suitable for both direct and sandwich ELISA formats. Optimize coating concentrations with the immunogen peptide.
  • All dilutions and conditions should be empirically determined for the specific experimental system.
