- Histone H4 is one of the four core histone proteins (H2A, H2B, H3, H4) that form the nucleosome octamer, wrapping ~147 bp of DNA and compacting the genome into chromatin.
- As a nucleosome core component, H4 plays a central role in regulating DNA accessibility for transcription, replication, repair, and maintaining chromosomal stability.
- In humans, H4 is encoded by 14 nearly identical genes (H4C1–H4C16) clustered at multiple chromosomal loci, producing a conserved 103-amino acid protein.
- H4 localizes exclusively to the nucleus and chromosomes, where it is subject to extensive post-translational modifications (acetylation, methylation, phosphorylation, citrullination, ubiquitination, etc.) that constitute the "histone code" to govern chromatin dynamics.
- Modifications on the N-terminal tail (aa 1–20) are critical for gene regulation; for example, acetylation at specific lysines correlates with active transcription, while methylation can signal either activation or repression depending on the residue.

Experimental Guidance and Technical Tips
- The immunogen targets amino acids 1–100, encompassing the N-terminal tail and globular domain; consider that N-terminal epitope recognition may be affected by dense post-translational modifications in some chromatin contexts.
- For Western blot (WB), a strong band is expected at ~11 kDa; include acid-extracted histone samples as positive controls to enrich for basic proteins.
- In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), chromatin compacted in heterochromatic regions may mask the epitope; gentle DNAse digestion or mild permeabilization protocols can improve detection.
- For chromatin immunoprecipitation (ChIP), validate the antibody’s ability to enrich modified vs. unmodified nucleosomes under cross-linking and native conditions, as epitope accessibility varies with chromatin compaction.
- Cross-reactivity with mouse and rat is expected given the high sequence conservation (human/mouse H4 are identical); however, always confirm specificity in the relevant species and application.
