Biological Background: Complement decay-accelerating factor (CD55) Function and Localization
- CD55, also known as decay-accelerating factor (DAF), is a 41kDa glycoprotein that belongs to the complement regulatory protein family and is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) moiety.
- Its primary function is to protect host cells from complement-mediated damage by binding to C4b and C3b fragments, preventing the formation and accelerating the decay of the C3 and C5 convertases, thereby inhibiting the complement amplification cascade. - CD55 serves as a receptor for several enteroviruses, including coxsackieviruses A21, B1, B3, B5, human enterovirus 70 and D68, and echoviruses 6, 7, 11, 12, 20, 21, facilitating viral entry into host cells.
- It is widely expressed on cell membranes of all cell types in contact with plasma complement proteins, including epithelial cells lining extracellular compartments, and is also present in a secreted form in body fluids.
- The CD55 protein contains four short consensus repeats (SCRs), also known as sushi domains, and features disulfide bonds that stabilize its structure; alternative splicing generates multiple isoforms.
- Genetic variants in CD55 are associated with CHAPLE syndrome (complement hyperactivation, angiopathic thrombosis, and protein-losing enteropathy), and the molecule carries the Cromer blood group antigen system.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 50-150 of human CD55, located in the N-terminal region; this region is extracellular and suitable for detecting both native and denatured protein.
- For Western blotting, CD55 runs at approximately 41kDa under reducing conditions; use appropriate human cell lysates (e.g., HeLa, Jurkat) as positive controls and validate the band with known inhibitors or siRNA knockdown.
- In immunohistochemistry (IHC-P), optimize antigen retrieval (e.g., heat-induced epitope retrieval) and primary antibody dilution; tissues with high CD55 expression such as kidney, lung, or vascular endothelium are recommended.
- For ELISA, this monoclonal antibody can be employed as a capture or detection antibody; perform checkerboard titrations to determine optimal coating and detection concentrations.
