Biological Background: CD55 Function and Localization
- Complement decay-accelerating factor (CD55/DAF) is a member of the regulators of complement activation (RCA) family and is also known by aliases CR, TC, DAF, CROM, and CHAPLE.
- It inhibits complement activation by binding C4b and C3b, blocking the formation of C4b2a and C3bBb convertases (PubMed:7525274) and destabilizing C3 and C5 convertases to prevent complement amplification (PubMed:28657829). Related references: PMID:7525274, PMID:28657829
- CD55 is expressed on the plasma membranes of all cell types exposed to complement, including epithelial linings, and also exists in secreted forms and within the extracellular matrix.
- The protein is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) moiety and contains multiple Sushi (CCP) domains, disulfide bonds, and N‑glycosylation sites; it carries the Cromer blood group system antigens.
- CD55 acts as a receptor for several viruses, including coxsackieviruses A21, B1, B3, B5, human enteroviruses 70 and D68, and echoviruses 6, 7, 11, 12, 20, and 21.
- The human CD55 gene (GeneID 1604) encodes this protein, and mutations are associated with CHAPLE syndrome (complement hyperactivation, angiopathic thrombosis, and protein-losing enteropathy).
Experimental Guidance and Technical Tips
- The immunogen is recombinant human CD55 protein; consider performing epitope mapping or using a blocking peptide to confirm specificity in your assay system.
- For Western blotting, a band corresponding to the core protein (~41 kDa) may be observed, but N‑glycosylation can increase the apparent molecular weight; validate in relevant cell lysates.
- In ELISA applications, optimization of coating antigen concentration and antibody dilution is recommended for best signal-to-noise ratio.
- Cross-reactivity with mouse CD55 has been reported; validate the antibody in the appropriate species' sample matrices if switching between human and mouse targets.
