Biological Background: CDC5L Function and Localization
- CDC5L (Cell division cycle 5-like protein), also referred to as pombe cdc5-related protein, CDC5, CEF1, PCDC5RP, and CDC5-LIKE, is a human protein encoded by the CDC5L gene (Gene ID: 988).
- It acts as a DNA-binding protein involved in cell cycle control and may function as a transcription activator.
- CDC5L plays a central role in pre-mRNA splicing as a core component of precatalytic, catalytic, and postcatalytic spliceosomal complexes.

PMID:28076346 - As a key subunit of the PRP19-CDC5L complex, it is required for activating pre-mRNA splicing and participates in the cellular DNA damage response (DDR).
Related references:
PMID:20176811 - CDC5L also functions within the minor spliceosome, contributing to the splicing of U12-type introns in pre-mRNAs.
- The protein localizes to the nucleus, nuclear speckles, and cytoplasm.
- CDC5L is a phosphoprotein and undergoes ubiquitination-like modifications (isopeptide bond, Ubl conjugation), as revealed by proteomic analyses.
- It is ubiquitously expressed in both fetal and adult tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide spanning residues 300-400 of human CDC5L; thus, the antibody is expected to detect total CDC5L protein.
- For Western blot, a specific band at approximately 92 kDa should be observed; consider using nuclear extracts or whole cell lysates from human or mouse samples.
- In IHC-P, nuclear and nuclear speckle staining is anticipated; optimize antigen retrieval conditions and validate with appropriate tissue controls.
- When performing ELISA, direct or sandwich formats can be employed; titration of the antibody and validation with the immunogen peptide are recommended.
