Biological Background: CD45 Function and Localization
- CD45, also known as leukocyte common antigen (L-CA) or T200, is a receptor-type tyrosine-protein phosphatase (PTPRC) that plays a critical role in immune cell signaling.
- It functions as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one. Upon T-cell activation, recruits and dephosphorylates SKAP1 and FYN. - CD45 interacts with CLEC10A at the antigen-presenting cell–T cell interface, contributing to the modulation of T-cell activation thresholds and preventing autoreactivity.
- Alternatively spliced isoforms (ranging from isoform 1 to 8) are differentially expressed in thymocytes, with isoforms 1,2,3,5,7 detected and others not. Multiple protein weights (131–147 kDa) reflect this isoform diversity.
