Biological Background: CD352/SLAMF6 Function and Localization
- SLAMF6 (CD352, NTB-A) is a self-ligand receptor of the signaling lymphocytic activation molecule (SLAM) family, modulating the activation and differentiation of NK, T, and B cells in both innate and adaptive immunity.
- In natural killer (NK) cells, SLAMF6 triggers cytolytic activity only when they express high surface densities of natural cytotoxicity receptors, signaling via VAV1 phosphorylation and requiring SH2D1B rather than SH2D1A. Related references: PMID:11489943, PMID:16920955
- SLAMF6 promotes T-cell differentiation into a Th17 phenotype, increasing IL-17 secretion; this costimulatory activity is dependent on SH2D1A. Related references: PMID:16920955, PMID:22184727
- Together with SLAMF1, SLAMF6 controls the transition between positive selection and the subsequent expansion and differentiation of the thymocytic natural killer T (NKT) cell lineage (by similarity).
- In the absence of SH2D1A/SAP, SLAMF6 can transmit negative signals to CD4+ T-cells and NKT cells, and it negatively regulates germinal center formation by inhibiting T-cell:B-cell adhesion (by similarity).
