  • AKR1C3, also known as 17-beta-hydroxysteroid dehydrogenase type 5, 3-alpha-HSD type II, DD3, PGFS, and other aliases, is encoded by the AKR1C3 gene in humans.
  • It is a cytosolic aldo-keto reductase that catalyzes NADPH-dependent reduction of ketosteroids to hydroxysteroids with broad substrate specificity, primarily generating 17beta-hydroxysteroids.

  • AKR1C3 plays a central role in androgen biosynthesis, operating in both the classical (converting delta4-androstenedione to testosterone) and backdoor (reducing androsterone to 5alpha-androstane-3alpha,17beta-diol) pathways.
