Biological Background: AKR1C1 Function and Localization
- AKR1C1 (Aldo-keto reductase family 1 member C1) is also known as 20alpha-hydroxysteroid dehydrogenase, dihydrodiol dehydrogenase 1, and high-affinity hepatic bile acid-binding protein, reflecting its multifunctional roles in steroid and xenobiotic metabolism.
- As a cytosolic NADPH-dependent oxidoreductase, AKR1C1 catalyzes the stereospecific reduction of ketosteroids to hydroxysteroids, preferentially generating 20alpha-hydroxysteroids, but also 3alpha/beta- and 17beta-hydroxysteroids, thus modulating the activity of various steroid hormones. - In neurosteroid metabolism, AKR1C1 converts 5alpha-dihydrodeoxycorticosterone into the neuroactive steroid 3alpha,5alpha-tetrahydrodeoxycorticosterone (allopregnanolone precursor), which potentiates GABA(A) receptors, and also inactivates 3alpha,5alpha-tetrahydroprogesterone, thereby influencing neuronal excitability.
