Biological Background: AKR1C4 Function and Localization

  • AKR1C4 (aldo-keto reductase family 1 member C4), also known as 3-alpha-hydroxysteroid 3-dehydrogenase type I (3-alpha-HSD1), chlordecone reductase (CDR), dihydrodiol dehydrogenase 4 (DD4), or HAKRA, is a cytosolic enzyme.
  • It catalyzes the NADPH-dependent reduction of a broad range of ketosteroids with distinct positional and stereospecificity, producing 3alpha/beta-, 17beta-, and 20alpha-hydroxysteroids, and it inactivates 5alpha-dihydrotestosterone and progesterone by converting them to androstanediols and (20S)-hydroxypregn-4-en-3-one, respectively.   • As part of the 'backdoor' androgen biosynthesis pathway, AKR1C4 cooperates with AKR1C2 to convert 5alpha-dihydroprogesterone to allopregnanolone, leading to 5alpha-dihydrotestosterone secretion essential for embryonic gonad differentiation into testis. Related references: PMID:21802064
