Biological Background: CCL3 Function and Localization
- CCL3, also known as MIP-1-alpha or G0S19-1, is a small (10 kDa) secreted cytokine belonging to the CC chemokine family.
- It exhibits inflammatory and chemokinetic properties by binding to chemokine receptors CCR1, CCR4, and CCR5.
- CCL3 is a potent HIV-suppressive factor produced by CD8+ T-cells; recombinant MIP-1-alpha inhibits replication of HIV-1, HIV-2, and SIV in a dose-dependent manner.
- It contains structural features typical of chemokines, including conserved disulfide bonds and a signal peptide for secretion.
- Subcellularly, CCL3 is localized to the extracellular space upon secretion.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide from the mature human CCL3 protein (amino acids 24–92), which spans the core chemokine domain.
- For Western blotting, consider using low molecular weight markers due to the protein's small size (~10 kDa) and optimize transfer conditions for small proteins.
- In ELISA applications, this polyclonal antibody may be used for capture or detection in sandwich formats; validate performance using recombinant CCL3 standards and biological samples such as cell culture supernatants.
- As a polyclonal antibody, lot-to-lot consistency should be verified for quantitative assays.
- Cross-reactivity with mouse and rat CCL3 allows for multi-species studies, but species-specific sequence differences may necessitate validation in the target sample matrix.
