Biological Background: MSH2 Function and Localization
- MSH2 is a core component of the post-replicative DNA mismatch repair (MMR) system. It forms two heterodimers: MutS alpha (with MSH6) and MutS beta (with MSH3), which recognize and bind to DNA mismatches and insertion-deletion loops (IDLs).
- MutS alpha (MSH2-MSH6) recognizes single base mismatches and dinucleotide IDLs, while MutS beta (MSH2-MSH3) handles larger IDLs up to 13 nucleotides. After binding, the heterodimer recruits MutL alpha to direct downstream repair, including strand discrimination, excision, and resynthesis.
- MSH2 recruits DNA helicase MCM9 to chromatin to unwind mismatch-containing DNA. ATP binding and hydrolysis act as a molecular switch, converting MutS alpha into a sliding clamp that diffuses along the DNA backbone. Related references: PMID:26300262
- In addition to MMR, MSH2 may participate in DNA homologous recombination repair and modulate UV-B-induced cell cycle regulation and apoptosis in melanocytes.
- MSH2 is localized in the nucleus and associates with chromosomes during DNA repair processes.
- The protein is ubiquitously expressed across human tissues and is associated with hereditary nonpolyposis colorectal cancer (HNPCC, Lynch syndrome). It functions as a tumor suppressor and undergoes post-translational modifications, including phosphorylation, acetylation, and ubiquitination.
- Alternative splicing generates multiple MSH2 isoforms.

- The immunogen spans amino acids 330–583, a conserved core domain region, which may account for the observed cross-reactivity with mouse and rat MSH2.
- For WB, a specific band at approximately 105 kDa is expected; KO validation confirms target specificity, so consider including MSH2 knockout lysate as a negative control.
- In IHC-P, nuclear staining should be anticipated; optimize antigen retrieval and antibody dilution for the tissue of interest.
- For ELISA, the antibody can serve as a capture or detection reagent; pairing with another MSH2-specific antibody may improve assay performance.
