Biological Background: HMGB2 Function and Localization

  • HMGB2 (High Mobility Group Protein B2), also known as HMG-2, is a multifunctional non-histone chromosomal protein.
  • In the nucleus, HMGB2 binds DNA with a preference for non-canonical structures like single-stranded DNA, bending DNA and enhancing flexibility to facilitate transcription, chromatin remodeling, and V(D)J recombination. Related references: PMID:11909973, PMID:18413230, PMID:19522541
  • It acts as a cofactor for the RAG complex in V(D)J recombination, stimulating cleavage and RAG protein binding at recombination signal sequences.
  • HMGB2 localizes to the nucleus, chromosome, and cytoplasm, and can be secreted; extracellularly, it acts as a chemokine promoting proliferation and migration of endothelial cells via AGER/RAGE. Related references: PMID:19811285
  • It participates in innate immunity by acting as a promiscuous DNA/RNA sensor and has antimicrobial activity in gastrointestinal epithelial tissues. Related references: PMID:23877675
  • The protein is subject to acetylation, phosphorylation, and disulfide bond formation, and its activity is modulated by redox state.
  • HMGB2 is expressed in gastric and intestinal tissues and is implicated in neurogenesis and articular cartilage maintenance via the LEF1/Wnt pathway.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-100) of human HMGB2; it is advisable to confirm that the antibody recognizes the full-length protein under experimental conditions.
  • For chromatin immunoprecipitation (ChIP) and CUT&Tag assays, consider optimizing crosslinking and shearing conditions as HMGB2 is a chromatin-associated protein.
  • Endogenous HMGB2 is detectable in human, mouse, and rat samples; choose appropriate loading controls and validate the antibody in your specific tissue or cell line.
  • For immunohistochemistry (IHC) and immunofluorescence (IF), test antigen retrieval methods to unmask epitopes in formalin-fixed paraffin-embedded sections.
