Biological Background: TLR2 Function and Localization

  • TLR2 (Toll-like receptor 2), also known as CD282 and TIL4, is a leucine-rich repeat-containing receptor with a TIR domain that forms disulfide bonds and undergoes glycosylation and ubiquitin-like conjugation.
  • TLR2 cooperates with LY96 and TLR1 or TLR6 to recognize bacterial lipoproteins and lipopeptides, initiating signaling via MYD88 and TRAF6 that activates NF-kB, promoting inflammatory cytokine secretion and possibly apoptosis.   • TLR2 forms ligand-dependent activation clusters with CD14 and CD36, such as TLR2:TLR6:CD14:CD36 for diacylated lipopeptides and TLR2:TLR1:CD14 for triacylated lipopeptides, which signal from the cell surface and traffic to the Golgi. Related references: PMID:16880211
