Biological Background: Moesin Function and Localization

  • Moesin (MSN) is an ERM family protein that tethers the actin cytoskeleton to the plasma membrane.
  • Its activity is regulated by phosphorylation of a C-terminal threonine, which induces an open conformation for F-actin binding and cytoskeletal rearrangement. Related references: PMID:10212266
  • Activated moesin controls cell shape, membrane trafficking, and signal transduction. Related references: PMID:12387735, PMID:15039356
  • In the immune system, moesin is essential for lymphocyte homeostasis, self-tolerance, and egress from lymphoid organs. Related references: PMID:9298994, PMID:9616160
  • It contributes to immunologic synapse formation and modulates phagolysosomal biogenesis in macrophages (By similarity). Related references: PMID:27405666
  • Subcellularly, moesin is found at the cell membrane, apical microvilli, cytoplasm, and associated with the cortical cytoskeleton.
  • It is expressed in all tissues and is subject to multiple post-translational modifications, including phosphorylation, acetylation, and S-nitrosylation.

Experimental Guidance and Technical Tips

  • For Western blot, a band at approximately 68 kDa may be observed; consider using whole-cell lysates from human, mouse, or rat samples based on cross-reactivity data.
  • In immunohistochemistry (paraffin), antigen retrieval conditions may need optimization; moesin is ubiquitously expressed, so most tissues can serve as positive controls.
  • For ELISA, the immunogen sequence (aa 400-500) may be used to assess direct binding; pairing with other clones is advisable for sandwich ELISA development.
  • Since the antibody recognizes the C-terminal region containing the regulatory threonine, it may detect both phosphorylated and non-phosphorylated forms; validation in the relevant sample system is recommended.
