Biological Background: MSH2 Function and Localization
- MSH2 is a central component of the DNA mismatch repair (MMR) system, essential for maintaining genomic stability by correcting base-base mismatches and insertion-deletion loops (IDLs) that arise during DNA replication.
- It forms two heterodimers: MutSα with MSH6, which recognizes single base mismatches and small (1–2 nucleotide) IDLs, and MutSβ with MSH3, which recognizes larger IDLs up to 13 nucleotides long.
- Upon mismatch binding, the MutS heterodimer bends the DNA and shields approximately 20 base pairs, subsequently recruiting the MutLα heterodimer (MLH1-PMS2) to form a ternary complex that directs downstream repair events, including strand discrimination, excision, and resynthesis.
- MSH2 recruits DNA helicase MCM9 to chromatin, which unwinds the DNA strand containing the mismatch to facilitate repair (PubMed:26300262).
