Biological Background: MSH2 Function and Localization

  • MSH2 encodes the DNA mismatch repair protein Msh2, also known as MutS protein homolog 2, a key component of the post-replicative DNA mismatch repair (MMR) system.
  • It forms two heterodimers: MutS alpha (MSH2-MSH6) that recognizes single base mismatches and small insertion-deletion loops, and MutS beta (MSH2-MSH3) that recognizes larger loops.
  • Upon DNA mismatch binding, the MutS complexes recruit MutL alpha and downstream factors, including DNA helicase MCM9, to unwind the DNA and initiate repair. Related references: PMID:26300262
  • MSH2 has ATPase activity that acts as a molecular switch, converting MutS alpha into a sliding clamp upon mismatch-triggered ADP-to-ATP exchange, enabling diffusion along DNA.
  • The protein is localized to the nucleus and associated with chromosomes, consistent with its role in DNA repair and genome maintenance.
  • It is ubiquitously expressed, reflecting its fundamental role in DNA repair across tissues.
  • Post-translational modifications include acetylation, phosphorylation, and ubiquitination, and it is subject to alternative splicing.
  • MSH2 is classified as a tumor suppressor, linked to hereditary nonpolyposis colorectal cancer (HNPCC), and involved in DNA damage response and homologous recombination repair.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide derived from amino acids 1–100 of human MSH2, located at the N-terminus; this antibody may recognize full-length MSH2 and N-terminal isoforms.
  • For western blot (WB), the predicted molecular weight of human MSH2 is 105 kDa; loading controls and molecular weight markers should be chosen accordingly.
  • Given cross-reactivity with mouse and rat, the antibody can be considered for studies across these species, but validation in the specific experimental system is recommended.
  • For ELISA applications, the antibody may be used as a capture or detection reagent; optimization of coating and detection conditions is advised.
