Biological Background: Sestrin-1 Function and Localization
- Sestrin-1 (PA26/SEST1) is a stress-inducible protein encoded by the SESN1 gene, with multiple isoforms produced by alternative splicing.
- Functions as an intracellular leucine sensor that negatively regulates the mechanistic target of rapamycin complex 1 (TORC1) signaling pathway. In leucine absence, it binds the GATOR2 subcomplex and inhibits TORC1; leucine binding disrupts this interaction, activating TORC1. Related references: PMID:25263562, PMID:26449471
- Plays a role in oxidative stress defense by positively regulating NFE2L2-dependent transcription of antioxidant genes through SQSTM1-mediated autophagic degradation of KEAP1. Related references: PMID:23274085
- May possess alkylhydroperoxide reductase activity via its N-terminal domain, contributing to the prevention of reactive oxygen species (ROS) accumulation (By similarity).
