Biological Background: Somatostatin Function and Localization
- Somatostatin, also known as growth hormone release-inhibiting factor, is a peptide hormone encoded by the SST gene.
- It primarily inhibits the secretion of multiple pituitary hormones, including growth hormone (GH1), prolactin (PRL), adrenocorticotropic hormone (ACTH), luteinizing hormone (LH), and thyroid-stimulating hormone (TSH).
- Beyond the pituitary, somatostatin may modulate glucagon release from pancreatic alpha cells, cardiac contractile function, and neuronal processes such as memory retention and hippocampal excitability.
- The protein is secreted and undergoes post-translational modifications including amidation, proteolytic cleavage at paired basic residues, and intramolecular disulfide bond formation.
- A study demonstrated that neuronostatin, a somatostatin-related peptide, selectively inhibits basal secretion of GH1 and LH, and potentiates somatostatin's inhibition of ghrelin-stimulated GH1 secretion, without affecting other pituitary hormones (PubMed:29615476). Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 23-116 of human preprosomatostatin, covering the full mature peptide region; the antibody may recognize multiple processed forms (e.g., somatostatin-14 and somatostatin-28).
- For IHC-P, consider optimizing antigen retrieval conditions, as the target is a small secreted peptide.
- In ELISA applications, the antibody may serve as a capture or detection reagent; pairing with another clone recognizing a distinct epitope is recommended for sandwich assays.
- Due to high sequence conservation across human, mouse, and rat, validate antibody specificity in each species using appropriate positive and negative controls.
- Initial titration experiments are advised to determine the optimal working dilution for each application and sample type.
