Biological Background: Alpha-1 Antitrypsin Function and Localization

  • Alpha-1-antitrypsin (SERPINA1), also known as alpha-1 protease inhibitor or serpin A1, is a 46.7 kDa secreted glycoprotein and a member of the serpin superfamily.
  • It functions primarily as an inhibitor of serine proteases, with elastase as its main target, and also inhibits trypsin, chymotrypsin, plasmin, thrombin, and plasminogen activator.
  • The aberrant form of SERPINA1 inhibits insulin-induced NO synthesis in platelets, decreases coagulation time, and exhibits proteolytic activity against insulin and plasmin.
  • A reversible chymotrypsin inhibitor isoform exists that inhibits elastase but not trypsin, and its major physiological role is protection of the lower respiratory tract against proteolytic destruction by human leukocyte elastase (HLE).
  • SERPINA1 is localized to the secretory pathway, including the endoplasmic reticulum, and is secreted into the extracellular space and extracellular matrix.
  • It is ubiquitously expressed, with known expression in leukocytes and plasma.
  • Post-translational modifications include glycosylation and phosphorylation; it is classified as an acute phase protein and plays roles in blood coagulation and hemostasis.
