Biological Background: SESN2 Function and Localization

  • SESN2 (Sestrin-2), also known as HI95, SES2, or SEST2, is a stress-inducible metabolic regulator belonging to the sestrin family.
  • Functions as an intracellular leucine sensor that negatively regulates mTORC1 signaling via the GATOR complex. In leucine absence, SESN2 binds GATOR2 to inhibit mTORC1; leucine binding disrupts this interaction, activating TORC1. • Mediates TP53-dependent inhibition of TORC1 signaling in response to genotoxic stress. Related references: PMID:18692468
  • Protects against oxidative stress by positively regulating NFE2L2-driven antioxidant gene expression through SQSTM1-dependent autophagic degradation of KEAP1. Related references: PMID:23274085
  • May reduce reactive oxygen species (ROS) levels via intrinsic alkylhydroperoxide reductase activity of its N-terminal domain. Related references: PMID:26612684
  • May suppress protein translation during endoplasmic reticulum stress by inhibiting mTORC1. Related references: PMID:24947615
  • Subcellular localization is primarily cytoplasmic, with wide tissue expression. Post-translational modifications include acetylation, phosphorylation, and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 211-480 of human SESN2, covering the C-terminal region. This design may aid in detecting full-length endogenous SESN2 in Western blot and ELISA applications.
  • Reported cross-reactivity with mouse suggests potential utility in rodent models; validate reactivity in your specific sample system.
  • For Western blot, consider using a suitable protein extraction method and loading control, as SESN2 may be induced under stress conditions.
  • Optimize antibody dilution and incubation conditions for your particular assay format and sample type.
