Biological Background: ADARB1 Function and Localization
- ADARB1 (double-stranded RNA-specific editase 1), also known as ADAR2, DRADA2, or RED1, is a member of the adenosine deaminase acting on RNA (ADAR) family that catalyzes the hydrolytic deamination of adenosine to inosine (A-to-I editing) in double-stranded RNA (dsRNA) substrates.
- A-to-I RNA editing by ADARB1 can modulate gene expression through multiple mechanisms, including alteration of codons and protein recoding, regulation of pre-mRNA splicing, modulation of RNA stability, and influence on microRNA biogenesis and targeting.
- ADARB1 is responsible for site-selective editing of key neuronal transcripts, such as glutamate receptors (GRIA2 at Q/R and R/G sites, GRIK2), serotonin receptor (HTR2C), GABA receptor (GABRA3), and potassium voltage-gated channel (KCNA1), resulting in amino acid changes that alter ion channel and receptor properties.
- The enzyme exhibits both editing-dependent and editing-independent proviral effects on HIV-1 replication: it directly edits the viral 5′UTR and suppresses EIF2AK2/PKR activation to enhance viral production.
- Alternative splicing and alternative promoter usage generate multiple isoforms; isoform 1 has lower catalytic activity, whereas isoform 2 displays higher catalytic activity in vitro.
- ADARB1 is primarily localized to the nucleus and nucleolus, consistent with its role in co-transcriptional and post-transcriptional RNA modification.
- Expression is highest in brain and heart, with lower levels in placenta, lung, liver, and kidney; in pediatric astrocytomas, decreased RNA-editing activity correlates with tumor malignancy grade.
- The protein contains double-stranded RNA-binding domains (dsRBDs) and a deaminase domain with a coordinated zinc ion, and it is regulated by phosphorylation. Disease associations include intellectual disability (NEDHYMS) and a role in innate antiviral immunity.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fragment corresponding to amino acids 180–380 of human ADARB1 (NP_001103.1). This region lies within the N-terminal catalytic/deaminase domain, and antibodies raised against it may detect the target in denaturing and native conditions; consider verifying reactivity in your specific sample type.
- Predicted molecular weight is 81 kDa; confirm migration by SDS-PAGE under reducing conditions. Post-translational modifications or isoform-specific variants may shift apparent molecular weight.
- For Western blot (WB), use appropriate positive controls such as human brain, heart, or cell lines known to express ADARB1. Cross-reactivity with mouse and rat orthologs permits use in rodent model studies.
- ELISA protocols may benefit from optimizing coating antigen and antibody titration. The polyclonal nature can provide sensitive detection in sandwich or indirect ELISA formats when paired with a validated monoclonal capture antibody.
