  • EDAR (Tumor necrosis factor receptor superfamily member EDAR) is a transmembrane receptor encoded by the EDAR gene. It is also known as DL, ED3, ED5, ED1R, EDA3, HRM1, EDA1R, ECTD10A, ECTD10B, and EDA-A1R.
  • It functions as a receptor for ectodysplasin A isoform A1 (EDA-A1), but not for EDA-A2. Upon ligand binding, it mediates activation of NF-kappa-B and JNK signaling pathways, and may promote caspase-independent cell death.
  • EDAR belongs to the tumor necrosis factor receptor superfamily and is crucial for the development of ectodermal tissues. Mutations in this gene are associated with ectodermal dysplasia syndromes, such as hypohidrotic ectodermal dysplasia.
  • Subcellularly, EDAR is localized to the membrane, consistent with its role as a cell surface receptor.
  • Tissue expression: Detected in fetal kidney, lung, skin, and cultured neonatal epidermal keratinocytes. It is not detected in lymphoblast and fibroblast cell lines.
  • Key post-translational modifications include glycosylation, and the protein contains a disulfide bond pattern characteristic of TNFR family members.
  • The protein exists in isoforms due to alternative splicing, and is involved in developmental processes and differentiation.

  • The immunogen corresponds to amino acids 27-187, which spans the extracellular domain of EDAR. This design likely generates antibodies recognizing the ectodomain, suitable for detecting membrane-bound receptor in whole cell lysates or tissue samples.
  • For Western blot, a band around 49 kDa is expected based on the calculated molecular weight; however, glycosylation may result in a higher apparent molecular weight. Consider using reducing and non-reducing conditions to assess disulfide-linked structures.
  • For ELISA, the antibody can be used for capture or detection of EDAR protein. Validate coating concentrations and detection ranges in your specific assay system.
  • Cross-reactivity is reported for human EDAR; testing with samples from other species should be validated individually.
