Biological Background: RAB1A Function and Localization
- RAB1A is a small GTPase of the Rab family that cycles between an inactive GDP-bound form and an active GTP-bound form, serving as a key regulator of intracellular membrane trafficking from the formation of transport vesicles to their fusion with membranes. Related references: PMID:20639577, PMID:20861236, PMID:21303926
- It specifically regulates vesicular protein transport from the endoplasmic reticulum (ER) to the Golgi compartment and onwards to the cell surface, and is involved in IL-8 and growth hormone secretion. Related references: PMID:21303926
- RAB1A is required to modulate the compacted morphology of the Golgi apparatus. Related references: PMID:26209634
- It regulates the level of the calcium-sensing receptor (CASR) present at the cell membrane. Related references: PMID:20861236
- RAB1A plays a role in cell adhesion and cell migration via its function in protein trafficking. Related references: PMID:20639577
- It is involved in autophagosome assembly and cellular defense reactions against pathogenic bacteria. Related references: PMID:22939626
- Subcellularly, RAB1A localizes to the Golgi apparatus, endoplasmic reticulum, early endosome, cytosol, membrane, and melanosomes.
- The protein undergoes various post-translational modifications including prenylation, acetylation, phosphorylation, and ubiquitination-like modifications, and exists in alternatively spliced isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human RAB1A, potentially recognizing both GDP- and GTP-bound forms. Consider relevant controls for activation state-specific detection.
- For Western blot, a band near 23 kDa is expected; post-translational modifications may cause slight shifts. Validate in the relevant sample system using recommended loading controls.
- In immunofluorescence/immunocytochemistry, expect staining of the Golgi apparatus and endoplasmic reticulum; co-localization with organelle markers can confirm specificity.
- Indirect ELISA may be used for quantification; optimize antibody dilution and consider antigen coating conditions for best performance.
- The antibody cross-reacts with mouse and rat, making it suitable for studies in these model organisms; verify cross-reactivity in your specific sample context.
