Biological Background: REG3A Function and Localization
- REG3A (Regenerating islet-derived protein 3-alpha), also known as HIP/PAP, is a secreted C-type lectin involved in antimicrobial defense and tissue regeneration.
- It exhibits bactericidal activity specifically against Gram-positive bacteria by binding to peptidoglycan carbohydrates (PubMed:16931762) and forming membrane-permeabilizing pores (PubMed:24256734). The protein also has bacteriostatic properties. Related references: PMID:16931762 PMID:24256734
- REG3A acts as a hormone, binding to the EXTL3 receptor to activate signaling pathways such as PI3K-AKT, regulating cell proliferation and differentiation in keratinocytes and pancreatic beta cells (PubMed:19158046, PubMed:22727489, PubMed:27830702). Related references: PMID:19158046 PMID:22727489 PMID:27830702
- The protein is highly expressed in keratinocytes, particularly in psoriatic skin (PubMed:22727489, PubMed:27830702), constitutively in the intestine, and is overexpressed in acute pancreatitis (PubMed:1469087). Related references: PMID:1469087 PMID:22727489 PMID:27830702
- Subcellularly, REG3A is secreted and contains disulfide bonds, as indicated by UniProt keywords, consistent with its extracellular antimicrobial and signaling functions.
- It belongs to the acute phase and inflammatory response pathways, playing a role in skin injury repair and pancreatic inflammation.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a recombinant immunogen corresponding to amino acids 27-175 of human REG3A, which shares sequence homology with mouse and rat REG3A, supporting the observed cross-reactivity.
- For Western blot (WB), consider using a 1:500–1:2000 dilution and validate with a positive control tissue such as human pancreatic or intestinal lysates; a band near 19 kDa is expected.
- For ELISA, the antibody may be used as a capture or detection reagent; optimize coating concentration and secondary antibody dilution using standard checkerboard titration.
- Since REG3A is a secreted protein, cell culture supernatants or tissue homogenates may be suitable sample types; perform appropriate extraction controls.
