Biological Background: ABCE1 Function and Localization
-
ABCE1 is a nucleoside-triphosphatase that hydrolyzes ATP, GTP, UTP, and CTP, driving ribosome disassembly into free 60S and 40S subunits after translation termination or ribosome stalling.

It plays a critical role in the No-Go Decay (NGD) pathway by disassembling stalled ribosomes in cooperation with the Pelota-HBS1L complex.
Related references:
PMID:21448132 -
ABCE1 participates in mitochondrial quality control; upon mitochondrial damage, it is ubiquitinated by CNOT4, generating polyubiquitin signals that trigger mitophagy.
Related references:
PMID:29861391 -
As an RNase L inhibitor, it antagonizes the binding of 2-5A to RNASEL, thereby down-regulating the interferon-regulated antiviral 2-5A/RNASEL pathway.
Related references:
PMID:9660177, PMID:11585831, PMID:9847332 -
During viral infection, ABCE1 may act as a chaperone for HIV-1 capsid assembly and facilitates down-regulation of the 2-5A/RNASEL pathway in EMCV and HIV-1 infections.
-
The protein is primarily localized in the cytoplasm but is also found in mitochondria, consistent with its dual roles in translation and mitochondrial homeostasis.
-
ABCE1 is a 67 kDa member of the ATP-binding cassette (ABC) transporter family, containing conserved nucleotide-binding domains and a 4Fe-4S cluster.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 320–599 of human ABCE1, a region likely to generate antibodies that recognize the native folded protein with high specificity.
- For Western blot, a band of approximately 67 kDa is expected in human and mouse cell lysates. Optimize loading amounts and antibody dilution empirically.
- ELISA setups may benefit from coating with recombinant ABCE1 protein; consider both direct and capture formats.
- The antibody cross-reacts with mouse, making it suitable for translational studies in rodent models; validate in the relevant sample system.
- Given ABCE1’s role in ribosome recycling, consider using polysome profiling or puromycin incorporation assays to confirm functional impact if using the antibody for co-immunoprecipitation.
