Biological Background: BRD4 Function and Localization

BRD4 (Bromodomain-containing protein 4) is a chromatin reader that recognizes acetylated histones, playing a central role in epigenetic memory and transcription regulation. It remains associated with acetylated chromatin throughout the cell cycle and helps preserve chromatin architecture for postmitotic gene transcription. During interphase, BRD4 recruits the P-TEFb complex to promoters and distal enhancers, facilitating RNA polymerase II CTD phosphorylation and transcriptional elongation. It also mediates NF-kappa-B activation by binding acetylated RELA and regulates p53-dependent transcription.

An isoform of BRD4 functions as a chromatin insulator in the DNA damage response by recruiting condensin-2 to acetylated histones, thereby limiting H2A.X phosphorylation spreading.

BRD4 localizes predominantly to the nucleus and associates with acetylated chromatin and mitotic chromosomes. Its chromatin binding is regulated by histone acetylation status, influenced by signals such as JNK pathway activation.

BRD4 is ubiquitously expressed and subject to extensive post-translational modifications, including phosphorylation at multiple serine residues (e.g., S470, S484, S488, S492, S494, S498, S499, S503, S601, S1117, S1126, S1201, S1204) and acetylation at K1111. This antibody specifically detects acetylation at Lys332, a modification not yet fully characterized in UniProt but designed based on the immunogen peptide.
