Biological Background: CREB3L2 Function and Localization
- CREB3L2 (also known as BBF2H7) is a transcription factor involved in the unfolded protein response (UPR) localized to the endoplasmic reticulum (ER) membrane under basal conditions.
- Upon ER stress, CREB3L2 is transported to the Golgi and cleaved by S1P and S2P proteases, releasing its N-terminal domain to translocate to the nucleus and activate transcription of target genes.
- The protein plays a critical role in chondrogenesis by activating SEC23A transcription, promoting secretion of cartilage matrix proteins, and possibly regulating ER biogenesis-related genes.
- In neuroblastoma cells, CREB3L2 protects against ER stress-induced death and activates transcription by directly binding to the CRE site. Related references: PMID:17178827
- CREB3L2 is widely expressed, with highest levels in placenta, lung, spleen, and intestine, and shows nuclear localization following activation.
