Biological Background: BRSK1 Function and Localization
- BRSK1 (also known as SAD-B, hSAD1) is a serine/threonine-protein kinase belonging to the AMPK family, encoded by the BRSK1 gene on chromosome 19.
- It plays a key role in neuronal polarization by phosphorylating microtubule-associated proteins such as MAPT/TAU at Thr-529 and Ser-579, and by phosphorylating WEE1 at Ser-642 to downregulate its activity.
- BRSK1 regulates centrosome duplication by phosphorylating gamma-tubulin (TUBG1 and TUBG2) at Ser-131, promoting their translocation to the centrosome.
- The kinase is activated by STK11/LKB1-mediated phosphorylation and subsequently phosphorylates substrates including CDC25B, CDC25C, and RIMS1, linking it to cell cycle and synaptic vesicle regulation.
- It localizes to the cytoplasm, nucleus, centrosome, and synaptic vesicles, aligning with its roles in cell cycle, DNA damage response, and neurotransmitter release.
- BRSK1 is involved in the UV-induced DNA damage checkpoint by inhibiting CDK1 activity through phosphorylation and activation of WEE1 and inhibition of CDC25B and CDC25C.
- It is widely expressed, with highest levels in brain and testis, and protein levels remain constant throughout the cell cycle.
Experimental Guidance and Technical Tips
- For Western blotting (WB), the predicted band is at ~85 kDa; using a reducing gel and appropriate molecular weight markers is recommended.
- In immunohistochemistry (IHC-P), signal may be influenced by antigen retrieval method; testing both citrate- and EDTA-based buffers could be beneficial.
- Since the antibody is polyclonal, lot-to-lot consistency should be verified for critical quantitative assays.
- The immunogen corresponds to a region within the kinase domain and C-terminal; thus, the antibody may recognize both full-length and splice variants if the epitope is preserved.
- Cross-reactivity with mouse and rat is indicated; however, sequence homology in the immunogen region should be checked if using non-human samples.
