- BRD7 (Bromodomain-containing protein 7), also known as BP75, NAG4, or CELTIX-1, is a 75 kDa nuclear protein that contains a bromodomain and coiled-coil region, acting as both a transcriptional coactivator and corepressor.
- It participates in chromatin remodeling and modulates gene expression by binding to target promoters, increasing histone H3 acetylation at Lys-9 (H3K9ac).
- BRD7 activates the Wnt signaling pathway by negatively regulating GSK3B activity, leading to dephosphorylation at Tyr-216 in a DVL1-dependent manner.
- As a coactivator for TP53, BRD7 promotes TP53 acetylation at Lys-382, facilitates TP53 recruitment to target promoters, and is required for TP53-mediated cell-cycle arrest upon oncogene activation.
- It also acts as a corepressor, down-regulating transcription of specific genes, and recruits BRCA1 and POU2F1 to the ESR1 (estrogen receptor) promoter.
- BRD7 inhibits cell cycle progression from G1 to S phase, consistent with its tumor suppressor activity.
- Subcellularly, BRD7 is localized to the nucleus and associated with chromosomes.
