- Alternative names: 75 kDa bromodomain protein, Protein CELTIX-1, and also known as BP75, NAG4, SMARCI1.
- Functions as both transcriptional coactivator and corepressor, and may play a role in chromatin remodeling.
- Activates the Wnt signaling pathway by negatively regulating GSK3B kinase activity (dephosphorylation at Tyr-216) in a DVL1-dependent manner.
- Acts as a corepressor by binding target promoters, increasing histone H3 acetylation at Lys-9 (H3K9ac), and downregulating gene expression (e.g., ESR1 promoter).
- Coactivates TP53-mediated transcription, promoting TP53 acetylation at Lys-382 and efficient recruitment to promoters; required for TP53-mediated cell-cycle arrest.
- Inhibits cell cycle progression from G1 to S phase, consistent with tumor suppressor activity.
- Subcellular localization: predominantly nuclear, associated with chromosomes.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitin-like conjugation (isopeptide bond).
