Biological Background: Histone H3 Function and Localization
Histone H3 is a core component of the nucleosome, the fundamental repeating unit of chromatin. Together with histones H2A, H2B, and H4, it forms an octamer around which approximately 147 base pairs of DNA are wrapped, thereby compacting the genome and regulating access to DNA-templated processes. Histones play essential roles in transcription regulation, DNA repair, DNA replication, and chromosomal stability.

Human histone H3 exists as several variants, including H3.1, H3.2, H3.3, and the testis-specific H3.1t (encoded by H3-4/HIST3H3, UniProt Q16695). These variants are primarily nuclear and associate with chromosomes, where they undergo a multitude of post-translational modifications (PTMs) such as acetylation, methylation, phosphorylation, and ubiquitination. The pattern of these modifications, often referred to as the 'histone code', dictates chromatin states and governs gene expression.

Experimental Guidance and Technical Tips
