Biological Background: Tubulin alpha-1A Chain Function and Localization
Tubulin alpha-1A chain (TUBA1A, also known as TUBA3, B-ALPHA-1) is a major constituent of microtubules. Microtubules are dynamic cylindrical polymers composed of alpha- and beta-tubulin heterodimers that assemble into linear protofilaments. They grow by the addition of GTP-tubulin dimers at the plus end, forming a stabilizing cap; below the cap, GDP-bound dimers predominate due to the GTPase activity of alpha-tubulin. This dynamic instability is essential for cell division, intracellular transport, and structural support.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic acetylated peptide around K40; thus, the antibody is expected to specifically recognize the acetylated form of alpha-tubulin. For detection of endogenous acetylation, consider using deacetylase inhibitors (e.g., trichostatin A) in your experimental system to preserve the modification.
- Validated applications include WB, IF-P, and ELISA. For WB, a band around 50 kDa is expected. Due to high conservation, cross-reactivity with mouse and rat samples has been confirmed; however, validation in your specific model system is recommended.
- In immunofluorescence (IF-P), ensure appropriate fixation and permeabilization to preserve microtubule structures. Co-staining with a total alpha-tubulin antibody can help assess acetylation levels.
- As with any polyclonal antibody, lot-to-lot consistency may vary; include positive and negative controls (e.g., untreated vs. deacetylase-treated cells) in each experiment.
