Biological Background: MVP Function and Localization
- MVP is the major structural component of vault ribonucleoprotein complexes, which are involved in cargo transport and signal transduction. Related references: PMID:40691181
- MVP oligomerizes to form a cage-like vault structure that can encapsulate minor vault components such as PARP4. Related references: PMID:40691181
- Vault particles are thought to participate in nucleo-cytoplasmic transport, potentially shuttling cargo between the nucleus and cytoplasm. Related references: PMID:15133037
- MVP down-regulates interferon-gamma (IFNG)-mediated STAT1 signaling and subsequent JAK activation, modulating immune signaling pathways. Related references: PMID:16418217
- It also negatively regulates SRC tyrosine kinase activity and downstream MAP kinase signaling cascades, influencing cell proliferation and survival. Related references: PMID:16441665
- Subcellularly, MVP localizes to the cytoplasm, nucleus, nuclear pore complex, and perinuclear region, consistent with its transport functions.
- MVP is expressed in most normal tissues, with higher levels in secretory epithelial cells and cells exposed to xenobiotics. Overexpression is frequently observed in multidrug-resistant cancer cells, highlighting its role in chemoresistance.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination, as annotated in UniProt.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1–100 of human MVP, corresponding to the N-terminal region. The antibody is likely to recognize an epitope within this region; consider using peptide competition or truncation mutants to confirm specificity.
- For Western blot, MVP migrates at approximately 99 kDa under reducing conditions. Use whole cell or tissue lysates from human, mouse, or rat sources. A single band near 99 kDa is expected; additional bands may reflect post-translational modifications or isoforms.
- For ELISA, the antibody can be used for direct detection or in a sandwich pair with a validated coating antibody. Optimize coating and detection antibody concentrations for maximum signal-to-noise ratio.
- Validate antibody performance in your specific sample system and application, as conditions may vary. Include appropriate positive (e.g., HeLa or multidrug-resistant cell lines) and negative controls.
