Biological Background: Rac1 Function and Localization
- Rac1 is a plasma membrane-associated small GTPase that acts as a molecular switch, cycling between active GTP-bound and inactive GDP-bound states.
- In its active state, Rac1 binds effector proteins to regulate phagocytosis, epithelial cell polarization, neuron adhesion, migration, differentiation, membrane ruffling, and osteoclast ruffled border formation.

Experimental Guidance and Technical Tips
- The recombinant immunogen covers amino acids 1-189 of human RAC1, a region highly conserved among Rac family members; thus, the antibody recognizes Rac1, Rac2, and Rac3.
- For Western blot, expect a band near 21 kDa. Validate in the relevant sample system and consider appropriate controls for active GTP-bound vs. inactive GDP-bound Rac.
- For IHC-P, optimize antigen retrieval conditions and validate staining in appropriate tissue sections known to express Rac proteins.
- For ELISA, the antibody can be explored as a capture or detection reagent; assay conditions should be optimized individually.
- Rac1 undergoes prenylation; ensure sample preparation maintains membrane localization if studying native interactions.
