Biological Background: Stathmin Function and Localization
- Stathmin (also known as Oncoprotein 18, Op18, p18, p19, and metablastin) is a ubiquitous cytosolic phosphoprotein that regulates microtubule dynamics by promoting depolymerization and preventing polymerization.
- Encoded by the STMN1 gene, stathmin exists in multiple isoforms and is a member of the stathmin family, characterized by a conserved coiled-coil domain.
- Phosphorylation at multiple sites tightly regulates its activity; phosphorylation at Ser-16 may be required for axon formation during neurogenesis (by similarity).
- Beyond neurogenesis, stathmin is involved in the control of learned and innate fear (by similarity).
- Subcellularly, stathmin localizes to the cytoplasm and associates with the cytoskeleton, consistent with its role in microtubule destabilization.
- Expression is highest in fetal and adult brain, spinal cord, and cerebellum, and is significantly elevated in acute leukemia cells compared to normal peripheral blood lymphocytes.
