Biological Background: SRC Family Tyrosine Kinases Function and Localization
- Proto-oncogene tyrosine-protein kinase Src (c-Src) is a non-receptor tyrosine kinase activated by diverse receptors (immune receptors, integrins, RTKs, GPCRs, cytokine receptors) to phosphorylate downstream effectors, regulating gene transcription, immune response, cell adhesion, apoptosis, and migration (PubMed:34234773). Related references: PMID:34234773
- At focal adhesions, Src phosphorylates key proteins such as PTK2/FAK1 and paxillin, linking integrin signaling to cytoskeletal reorganization and cell motility (PubMed:21411625). Related references: PMID:21411625
- Src participates in the RAS signaling pathway via phosphorylation of RASA1 and RASGRF1 (PubMed:11389730) and modulates EGF-dependent calcium-activated chloride channels (PubMed:18586953). Related references: PMID:11389730, PMID:18586953
- In the nucleus, Src may regulate cell cycle progression through interactions with nuclear proteins (PubMed:7853507) and supports anchorage-independent cell growth (PubMed:19307596). Related references: PMID:7853507, PMID:19307596
- Src is essential for osteoclast function and bone resorption; it forms a complex with PTK2B/PYK2 to phosphorylate CBL (PubMed:14585963, PubMed:8755529) and activates mitochondrial cytochrome C oxidase for energy production (PubMed:12615910). KISS1R signaling recruits Src and DUSP18 to dephosphorylate and inactivate Src, suppressing bone resorption (PubMed:38346942). Related references: PMID:14585963, PMID:8755529, PMID:12615910, PMID:38346942
- Src enhances antiviral innate immunity by phosphorylating components of the RIG-I pathway (PubMed:19419966) and phosphorylates various substrates including PDPK1, BCAR1, and CBLC to modulate diverse signaling outputs. Related references: PMID:19419966
- Localized to the cell membrane, mitochondrial inner membrane, nucleus, cytoplasm, cytoskeleton, and cell junctions (focal adhesions).
- Expressed ubiquitously, with highest levels in platelets, neurons, and osteoclasts (PubMed:22294297 supports expression in skin).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide encompassing the conserved Y416 activation loop site of the SRC family. For WB, a band around 60 kDa is expected.
- Due to high sequence conservation, the antibody is predicted to recognize multiple SRC family members (SRC, FYN, YES1, LYN, HCK, LCK) and cross-react with mouse and rat orthologs. Validate specificity with appropriate blocking peptides and phosphatase treatment controls.
- Consider using phospho-specific and pan-SRC antibodies to confirm phosphorylation status. For ELISA, the antibody may be used for phospho-peptide detection.
