  • MET, also known as Hepatocyte growth factor receptor (HGFR), proto-oncogene c-Met, and Scatter factor receptor, is a receptor tyrosine kinase that transduces signals from the extracellular matrix upon binding to its ligand HGF. It regulates key processes such as proliferation, scattering, morphogenesis, and survival.
  • Upon ligand binding, MET undergoes autophosphorylation at tyrosine residues including Y1234 and Y1235 in the activation loop, which creates docking sites for downstream effectors like PI3K, PLCG1, SRC, GRB2, STAT3, and GAB1. This triggers multiple cascades: RAS-ERK (morphogenetic effects), PI3K-AKT (prosurvival), and PLCgamma-PKC pathways.
  • During embryonic development, MET signaling is essential for gastrulation, neuronal precursor migration, angiogenesis, kidney formation, and skeletal muscle development (muscle progenitor migration and secondary myoblast proliferation). In adults, it contributes to wound healing, organ regeneration, tissue remodeling, and hematopoiesis.
  • MET is primarily localized to the cell membrane, but a secreted form also exists. It is expressed in normal hepatocytes and epithelial cells lining the stomach, small and large intestines, as well as in basal keratinocytes of esophagus and skin. Experimental Guidance and Technical Tips

  • The immunogen used for this antibody is a synthetic phosphorylated peptide spanning Y1234 and Y1235 of human MET. For WB and ELISA, consider including non-phosphopeptide competition or phosphatase-treated lysates to confirm phosphorylation specificity.
  • For WB, a band at approximately 156 kDa is expected. Since MET is a membrane protein, use appropriate lysis buffers (e.g., RIPA) and include protease/phosphatase inhibitors. For ELISA, validate the antibody with coated phospho-peptide and appropriate controls.
  • Stimulating cells with HGF before lysis can enhance detection of phosphorylated MET at these sites. Be aware of basal phosphorylation levels in your cell line of interest.
