- This antibody was raised against a synthetic phosphopeptide corresponding to the S1981 phosphorylation site of human ATM; specificity is expected for the phosphorylated form only.
- For Western blotting, include phosphatase inhibitors in lysis and wash buffers to preserve phosphorylation. Consider verifying phospho-specificity by treating samples with lambda protein phosphatase or by performing peptide blocking experiments.
- Due to the large size of ATM (~351 kDa), use a low-percentage acrylamide gel (e.g., 6%) and extended transfer times. A gradient gel (4-15%) may also improve separation.
- Validate the antibody in your specific model system; cross-reactivity with mouse is reported, but performance in other species should be tested individually.
- For ELISA, confirm coating and detection conditions, as phospho-specific antibodies may require optimized buffer pH and blocking reagents.
