Biological Background: Somatostatin (SST) Function and Localization
- Somatostatin (SST), also known as growth hormone release-inhibiting factor, is a peptide hormone encoded by the SST gene.
- It inhibits the secretion of numerous pituitary hormones, including growth hormone (GH1), prolactin (PRL), ACTH, luteinizing hormone (LH), and TSH. The inhibition of ghrelin-stimulated GH1 secretion can be further enhanced by neuronostatin, a peptide derived from the same precursor.
- Somatostatin may potentiate low-glucose-induced glucagon release from pancreatic alpha cells via GPR107 and PKA activation, and it has been implicated in regulating cardiac contractility, memory retention, hippocampal excitability, anxiety, feeding behavior, and arterial pressure (By similarity).
- Neuronostatin, another cleavage product of the SST precursor, selectively inhibits basal secretion of GH1 and LH without affecting other pituitary hormones (PRL, ACTH, FSH, TSH) and potentiates the inhibitory action of somatostatin on ghrelin-stimulated GH1 release. - The protein is secreted and undergoes post-translational modifications including amidation, disulfide bond formation, and cleavage on pairs of basic residues.
- The mature somatostatin peptide has a molecular weight of approximately 12.7 kDa; the recombinant immunogen yields an antibody recognizing a band around 13 kDa.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 23–116 of the human SST precursor, which includes the signal peptide and pro-region; the polyclonal antibody may therefore recognize both the proprotein and mature somatostatin forms. For Western blot, consider running samples under reducing and non-reducing conditions to distinguish monomeric and disulfide-linked species.
- Given the secreted nature of somatostatin, enhance intracellular detection in IF and IHC by using a secretion inhibitor (e.g., brefeldin A) or focusing on tissues with high peptide turnover.
- For ELISA assays, the polyclonal format provides broad epitope coverage; consider pairing this antibody with a monoclonal antibody for a sandwich ELISA setup after appropriate validation.
